Selection of Astaxanthin-Overproducing Mutants of Phaffia rhodozyma with beta-Ionone.
beta-Ionone, an end ring analog of beta-carotene, inhibits astaxanthin production in the red yeast Phaffia rhodozyma. Astaxanthin-overproducing mutants of this yeast are easily spotted on beta-ionone-containing yeast malt agar plates. beta-Ionone appears to block astaxanthin synthesis at the beta-carotene level.
متن کاملAstaxanthin production by Phaffia rhodozyma using a pH-stat control
Fed-batch essays using yeast Phaffia rhodozyma were performed to obtain high astaxanthin concentrations. The yeast was grown over an inexpensive media obtained from the Yucca fillifera date. Feeding was carried out through implantation of a pH-stat control system. Biomass (28.7 mg/l) and pigment (18 mg/l) concentration three folded those that were obtained by previous batch cultures.

A minimal medium which allowed the sustained, rapid growth of Zymomonas mobilis and the isolation of a range of auxotrophic mutants was developed.
متن کاملIsolation and characterization of mutants of Saccharomyces cerevisiae auxotrophic and conditionally auxotrophic for 5'-dTMP.
An improved method for isolation of yeast m utants auxotrophic for 5'-dTM P is presented. The procedure employs the two folic acid antagonists aminopterin and sulfanilamide (SA A). Selec tiveness of the procedure depends on concentration of SAA and time of incubation. 44 mutants auxotrophic and 3 conditionally auxotrophic for 5'-dTMP were isolated. All belong to one complementation group. The ...
متن کاملImpact of gamma rays on the Phaffia rhodozyma genome revealed by RAPD-PCR
BACKGROUND AND OBJECTIVES Phaffia rhodozyma is a red yeast which produces astaxanthin as the major carotenoid pigment. Astaxanthin is thought to reduce the incidence of cancer and degenerative diseases in man. It also enhances the immune response and acts as a free-radical quencher, a precursor of vitamin A, or a pigment involved in the visual attraction of animals as mating partners. The impac...
